Ipamorelin: Selectivity as a Design Goal

Ipamorelin is usually described as the selective one. Here is what selectivity actually means in receptor terms — and why it is a chemistry claim, not a safety claim.

Ipamorelin is a pentapeptide — five residues — developed as a growth hormone secretagogue acting at the ghrelin receptor, formally the growth hormone secretagogue receptor 1a. Its reputation rests on one word that gets used loosely: selectivity.

What selectivity means here

Earlier secretagogues in the same family were reported in the literature to also raise cortisol and prolactin, because the receptors involved sit in overlapping signalling territory. Ipamorelin was characterised as producing growth hormone release with comparatively little effect on those other axes in the models tested.

That is a pharmacological observation from specific animal and cell studies. It says the molecule engages one receptor population more cleanly than its predecessors did. It does not say the compound is safe, and it is not a claim about human use.

The two-lever system

Growth hormone release is governed by two separate inputs: GHRH acting on its own receptor, and ghrelin acting on GHSR-1a. They are different levers on the same machine. This is why the literature so often studies a GHRH analogue and a ghrelin-receptor agonist together — they probe complementary halves of the same axis.
